Serine/threonine protein phosphatase 2A (PP2A) dephosphorylates CARMA1 and negatively regulates TCR-induced NF-κB activation [16]. In addition, ubiquitination of CARMA1 also plays a role in altering its functions. Monoubiquitination of CARMA1 by E3 ubiquitin ligase casitas B-lineage lymphoma b (Cbl-b) disrupts its association with BCL10, and thus inhibits TCR-induced NF-κB activation [17]. Furthermore, TCR-activated CARMA1 undergoes lysine 48 (K48)-linked polyubiquitination and proteasomal degradation, which is an intrinsic negative feedback control mechanism to balance lymphocyte activation [18]. In an effort to understand the subtle mechanisms of T-cell activation, we previously endeavored to identify novel proteins participating in TCR signaling. By biochemical affinity purification, we identified a CARMA1-associated E3 ubiquitin ligase, stress-induced-phosphoprotein 1 homology and U-box containing protein 1 (STUB1, also known as CHIP) [19].
