Cytokines released upon antigen stimulation were measured using standardized methods: TGFβ and IL-17 by ELISA (Quantikine) and, 11 additional Th1 and Th2 cytokines (IL-1α/IL-1β/IL-2/IL-4/IL-5/IL-6/IL-8/IL-10/IL-12/IL-13/TNF-α) using multiplex cytokine array (Endogen). IHLs were stimulated with media or HCV-Core pool-1, pool-2, or pool-3 peptides in the presence of autologous B-LCL. Fibrogenic/fibrolytic effects of conditioned supernatants (dilution 1:20) from these stimulated IHL or direct coculture were assessed using human LX-2 HSC30 cultured in Dulbecco’s modified Eagle’s medium (DMEM) plus 2% fetal bovine serum (FBS) in triplicate.
